چکیده

The neurotransmitter dopamine has been implicated in the aetiology of brain disorders such as Parkinson’s disease and schizophrenia, and dopamine receptors provide important targets for anti-parkinsonian and antipsychotic drugs. In particular, the beneficial use of dopamine D2 or DI/D2 agonists in the drug treatment of Parkinson’s disease is now well established [l], although the ideal agonist has not yet been identified. Alternative strategies for the treatment of this condition which are currently being evaluated include the use of D2 partial agonists alone or in combination with a full-efficacy DI agonist [2]. Another proposed therapeutic application for D2 receptor partial agonists is in the treatment of psychostimulant addiction [3]. Hence, elucidation of the molecular mechanisms underlying drug-receptor interaction and response and a simple procedure for predicting agonist intrinsic activity in vitm would facilitate the design and development of new agents with therapeutic potential.

Effects of selective dopamine receptor (Dl and D2) agonists on stress-induced gastric ulcer in rat

It has been revealed that dopamine (DA) modulates gastro-duodenal responses to stress. Several investigations have been made to identify the mechanisms and/or receptors by which DA or its promoters exert their gastroprotective function against stress, however there are many discrepancies in this respect. In order to clarify the relative contribution and/or interaction of two DA receptor subty...

Selective Inhibitory Effect of Adenosine A1 Receptor Agonists on the Proliferation of Human Tumor Cell Lines

Background: In this study, the effects of three structural analogues of adenosine upon proliferation of human tumor cells were investigated. Previous research showed a cytotoxic effect of adenosine via A3 receptor and A1 receptor and sometimes this effect was receptor independent. The researches showed a differential cytotoxic effect of adenosine and its A3 agonists on cancerous cells, while ot...

Agonist action at D2(long) dopamine receptors: ligand binding and functional assays.

1. The activities of a range of agonists at D2(long) dopamine receptors expressed in CHO cells have been determined in ligand binding and in a functional assay, the stimulation of [35S]-GTPgammaS binding. 2. For several agonists (apomorphine, dopamine, pergolide, quinpirole, NPA, ropinirole, talipexole) binding in the absence of added guanine nucleotides was best described in terms of interacti...

Presence of prejunctional D2-dopaminoceptors and α2-adrenoceptors on the cholinergic nerve of the common bile duct of guinea pig

On most adrenergic and cholinergic nerve terminals, prejunctional α-adrenoceptors belonging to the α2-subtype have been identified. Activation of these receptors will decrease the release of norepinephrine. It has been reported that several isolated tissue preparations contain prejunctional dopamine receptors, the stimulation of which inhibits neurotransmission. It has remained uncertain whethe...

EFFECTS OF CCK RECEPTOR AGONISTS AND ANTAGONISTS ON MORPHINE-INDUCED ANTINOCICEPTION IN MICE

In the present study the effects of both CCK receptor agonists and antagonists on antinociception induced by morphine in the tail-flick test have been evaluated. M orphine induced dose-dependent antinociception in mice. The response of morphine was potentiated by sulfated cholecystokinin-8 (CCK-8S) but not by unsulfated cholecystokinin-8 (CCK-8U). The CCK receptor antagonists MK-329 and L-...
